General anaesthetics and bacterial luminescence. I. The effect of diethyl ether on the in vivo light emission of Vibrio fischeri.

چکیده
The factors which determine the sensitivity of bacterial luminescence to inhibition by the general anaesthetic, diethyl ether, have been investigated. The in vivo luminescent reaction of Vibrio fischeri displays a change in sensitivity to this agent during the bacterial growth cycle. This variation is particularly marked during the lag phase of growth and detailed investigations of the effect of potassium cyanide and w-decanal on the potency of ether during this early period are described. The results suggest th a t fluctuations in the substrate levels available to the light-producing enzyme are responsible for the variation in sensitivity to ether.

In silico structural analysis of quorum sensing genes in Vibrio fischeri
Quorum sensing controls the luminescence of Vibrio fischeri through the transcriptional activator LuxR and the specific autoinducer signal produced by luxI. Amino acid sequences of these two genes were analyzed using bioinformatics tools. LuxI consists of 193 amino acids and appears to contain five α-helices and six ß-sheets when analyzed by SSpro8. LuxI belongs to the autoinducer synthetase fa...
متن کاملMyristic acid stimulation of bacterial bioluminescence in "aldehyde" mutants.
The involvement of long chain aldehyde in bacterial luminescence was known both from its being required for light emission in the in vitro reaction with pure luciferase and from its ability to stimulate luminescence in vivo in a certain class of dark "aldehyde" mutants. We have found that the luminescence of some (but not all) of such aldehyde mutants is also stimulated by long chain aliphatic ...
متن کاملInduction of light emission by luminescent bacteria treated with UV light and chemical mutagens.
Intensity of light emission by luminescent bacteria in response to UV irradiation and chemical mutagens was tested. We demonstrated that luminescence of six strains of marine bacteria (belonging to four species: Photobacterium leiognathi, P. phosphoreum, Vibrio fischeri and V. harveyi) is significantly increased by UV irradiation relatively shortly after dilution of cultures. Such a stimulation...
متن کاملLitR, a new transcriptional activator in Vibrio fischeri, regulates luminescence and symbiotic light organ colonization.
Vibrio fischeri is the bacterial symbiont within the light-emitting organ of the sepiolid squid Euprymna scolopes. Upon colonizing juvenile squids, bacterial symbionts grow on host-supplied nutrients, while providing a bioluminescence that the host uses during its nocturnal activities. Mutant bacterial strains that are unable to emit light have been shown to be defective in normal colonization....
متن کاملEffects of a pre-incubation period on the photoinduced toxicity of polycyclic aromatic hydrocarbons to the luminescent bacterium Vibrio fischeri.
Irradiation of polycyclic aromatic hydrocarbons (PAHs) in aqueous solution with simulated solar radiation (SSR; a light source with a visible light: UV-A:UV-B ratio similar to that of sunlight) can greatly enhance their toxicity. Two microbial toxicity tests with Vibrio fischeri were used to investigate the effect of composition of the growth medium and pre-incubation on the photoinduced toxici...
